A Szamár-kő a Mátrában az Ágasvár kettős csúcsától keletre található sziklaalakzat, mely a Newton-sziklával együtt vulkáni tevékenység során jött létre. A hatalmas képződmény szemmel láthatóan kettős osztatú: alsó része, „talapzata" keskenyebb, mint a tetején buzogányszerűen kiszélesedő felső egység. A válogató erózió a puhább kőzetek lepusztításával kitakarta a sziklát, később pedig kikezdte annak kevésbé ellenálló alapját - ezért a buzogány alak. A folyamat most is tart, egy ponton pedig a nehéz, keményebb, lassabban pusztuló fölső részt nem fogja elbírni a gyorsabban vékonyodó nyak, a hatalmas szikla nagy robajjal görög majd le a meredek lejtőn.
Alsó része finomabb szemű, vulkáni-üledékes sorozat, melyre a jóval keményebb, láva által cementált törmelékkő (breccsa) települt, ez magyarázza a kőzetminőség eltérését és a figyelemfelkeltő alakot. A lejjebb megtalálható tufák (a kitörésekkor aláhullot törmelékes kőzetek) és a rájuk támaszkodó, lávától összesült összlet a rétegvulkán periodikus (hol lávaöntő, hol robbanásos) működésének hírmondói.
Ha hasonló keletkezésű sziklaalakzatokat szeretnénk megfigyelni, nem kell messze mennünk: a Csörgő-patak völgyében megkereshetjük a Bárány-követ vagy a Csóka-követ is.

- A buszmegállóból a K jelzéseket követve nyugati irányba induljunk a Petőfi Sándor utcán, majd térjünk jobbra a Virág utcára, ami kivezet a településről. 2,9 km kellemesen emelkedő séta után forduljunk a meredeken jobbra kaptató K▲ és ʍ jelzésekre, amik a Newton-szikla érintésével a Szamár-kőhöz vezetnek (összesen 3 km).

- Mátraszentistván központjában tágas, ingyenes parkolóban tudjuk hagyni az autónkat.
