Der Dom Santa Maria Assunta wurde ab dem Jahr 1175 in Spoleto gebaut, nachdem der Vorgängerbau von den Truppen Friedrich Barbarossas zerstört worden war. Der Dom ist ein Beispiel romanischer Baukunst. Die Fassade ist nicht einheitlich, da sie im Laufe der Jahre erweitert wurde. Acht Fensterrosen sorgen für die Beleuchtung des Innenraums, der 1638 komplett barockisiert wurde. Der Dom hat mit den umliegenden Gebäuden zusammen eine großartige Wirkung und wird als Theaterschauplatz genutzt. Beispielsweise Aufführungen des Festivals dei Due Mondi werden vor der Domfassade gezeigt. Simon von Collazzone, ein Gefährte des heiligen Franziskus liegt im Dom begraben.
