Ursprünglich handelt es sich bei der Synagoge um eine Katharinenkirche aus dem Jahr 1419. Geplagt von zahlreichen Bränden wurde sie 1714 mit dem Einzug der Hugenotten als einschiffiger Putzbau mit Walmdach und Sakristeianbau errichtet. Ihren Turm im neugotischen Stil erhielt die Cottbuser Kirche 1870. 2003 wurde die Schlosskirche renoviert und die Innenausstattung dem architektonischen Stil nachempfunden.
Nach der Zerstörung der Synagoge in der Karl-Liebknecht-Str. im Jahr 1938 und dem Ende des Holocaust wurde das Gebäude als jüdisches Gotteshaus zur Verfügung gestellt. Mit Ausstellung, zum Beispiel über die Geschichte der Schlosskirche, ist die Synagoge der gesamten Öffentlichkeit zugänglich geblieben.
