Jako jediná ze synagog se ta v Třešti vyznačuje podloubím u vstupu. S budovou je spojován také Franz Kafka, který tu u svého strýce trávil po sedm let letní prázdniny.
Třešťská synagoga se nacházela v jižní části židovského ghetta. Jeho přesná doba vzniku se neví, ale v roce 1825 ji zničil požár, takže museli postavit novou v empírovém stylu. Má klenutý sál se štukovou výzdobou a průčelí s podloubím v klasicistním stylu. Je to jedné svého druhu v Česku. Interiér se bohužel nedochoval. Od roku 1957 sloužila budova Církvi československé husitské, která tu stále koná bohoslužby. Ještě o miléniu tam byly vystavené betlémy.
Dnes se v synagoze konají koncerty a výstavy. V přízemí mohou návštěvníci vidět stálou expozici Zaváté stopy, která mapuje historii židovské obce v Třešti. V prvním patře je výstava věnovaná životu Franze Kafky, který v letech 1900 – 1907 zde pobýval u svého strýce Siegfrieda Löwyho a pravděpodobně tu vznikl i jeho román Venkovský lékař. Konají se tu pravidelné prohlídky s průvodcem.
